Catalytic autoimmune antibodies from the sera of lupus patients were purified using histidyl-aminohexyl-Sepharose gel and compared with the antibodies purified with protein A and protein G affinity chromatography. The IgG preparations from the histidine affinity column had a much higher catalytic activity in hydrolyzing the peptide substrate Pro-Phe-Arg-methyl-coumarinamide compared to the antibodies obtained by the conventional protein A/G method. This preservation of catalytic activity is attributed to the gentle buffer conditions used in the histidine ligand method that allowed the integrity of three-dimensional structure of purified catalytic antibodies. Thus, histidine affinity offer a superior method for isolating autoimmune catalytic antibodies.  相似文献

Phenolic compounds are a sort of common pollutants in water. Phenol index becomes an expedite indicator for the evaluation of the contamination level of water samples, in spite of the knowledge of the individual phenol and its derivatives are also important.In this work, an environmental friendly method for the determination of phenols, using a segmented flow system based on the conventional method's reactions without the liquid-liquid extraction step, was validated.Three linear dynamic ranges using C6H5OH: 1-10 μg l−1, 10-200 μg l−1 and 0.2-2.5 mg l−1, with a coefficient of variation lower than 2%, were obtained. Several method's performance parameters were determined: limits of detection, limits of quantification, precision through duplicate analysis and trueness using the reference materials purchased from LGC Promochem, RTC no. QCI-043-2 Lot:P1. Measurement uncertainty was evaluated using an interlaboratory approach based on proficiency testing data. Relative combined uncertainty for phenols in water samples, , of 0.054 were obtained, in according to those imposed by the Portuguese Legislation: target for 1 μg l−1 of phenol (surface waters) and target for 500 μg l−1 of phenol (wastewaters).A high efficiency reduction and elimination of reagents and wastes, reduction of analysis time and exposition of the analyst were also obtained.  相似文献

Despite an increasing focus on low level methods for determination of mercury species in water over the last decades, few studies have paid attention to direct effects of different sample preparation methods (i.e. preservation techniques) on natural freshwater samples. In this study we show how different preservation techniques give significantly different concentrations of total and methylmercury in freshwaters (9 and 14% on average, respectively). Natural stream samples from a forested lake catchment were studied. Mean stream sample concentrations of total (3.6 ng/L) and methylmercury (0.06 ng/L) reflect levels typical for pristine humic boreal catchments. The main reason for the observed average differences in total and methylmercury concentrations is the use of one instead of two sample bottles and timing of sample acidification, respectively.  相似文献

Time-resolved polarization-dependent kinetics of near infrared (NIR) spectral wing (SW) emission from human cancerous and normal prostate tissues was investigated. The differences between the temporal profiles, intensities and polarization properties of SW emission from cancerous and normal prostate tissues were observed. The intensity of SW emission from cancerous prostate tissue at peak position was found ∼2 times stronger than the normal prostate tissue indicating changes of fluorophores contained in the cancerous tissue compared with normal tissue. The decay time of anisotropy of SW emission from cancerous tissue was found ∼1.4 times longer than that of normal tissue reflecting changes of the micro-environment of fluorophores in the cancerous tissue as compared with the normal tissue. The stronger intensity of SW emission from cancerous tissue was used to image and identify cancerous areas surrounded by normal tissue.  相似文献

高超声速边界层转捩的准确预测对飞行器的防热、减阻至关重要,而影响高超声速边界层转捩的因素众多.从模式角度出发研究边界层转捩的影响因素,采用k-ω-γ 转捩模式对5°圆锥的边界层转捩进行了数值分析,计算了不同头部钝度、来流雷诺数和湍流度情况下的边界层转捩,并与实验结果进行了对比. 研究结果表明:k-ω-γ 转捩模式基本能够反映头部钝度、来流雷诺数、来流湍流度对高超声速圆锥边界层转捩的影响规律,但对转捩后的热流峰值预测不准;从模式构造角度分析发现,雷诺数越高或头部钝度越小,层流区边界层越薄,k-ω-γ 转捩模式中第一、第二模态时间尺度增大,因此转捩起始位置提前;来流湍流度越大,等效脉动动能初值越大,导致层流区发展过程中等效脉动动能越大,因此转捩易于发生.   相似文献

A discretization is presented for the initial boundary value problem of solidification as described in the phase-field model developed by Penrose and Fife (1990) [1] and Wang et al. (1993) [2]. These are models that are completely derived from the laws of thermodynamics, and the algorithms that we propose are formulated to strictly preserve them. Hence, the discrete solutions obtained can be understood as discrete dynamical systems satisfying discrete versions of the first and second laws of thermodynamics. The proposed methods are based on a finite element discretization in space and a midpoint-type finite-difference discretization in time. By using so-called discrete gradient operators, the conservation/entropic character of the continuum model is inherited in the numerical solution, as well as its Lyapunov stability in pure solid/liquid equilibria.  相似文献

We present a new unconditionally positivity‐preserving (PP) implicit time integration method for the DG scheme applied to shallow water flows. This novel time discretization enhances the currently used PP DG schemes, because in the majority of previous work, explicit time stepping is implemented to deal with wetting and drying. However, for explicit time integration, linear stability requires very small time steps. Especially for locally refined grids, the stiff system resulting from space discretization makes implicit or partially implicit time stepping absolutely necessary. As implicit schemes require a lot of computational time solving large systems of nonlinear equations, a much larger time step is necessary to beat explicit time stepping in terms of CPU time. Unfortunately, the current PP implicit schemes are subject to time step restrictions due to a so‐called strong stability preserving constraint. In this work, we hence give a novel approach to positivity preservation including its theoretical background. The new technique is based on the so‐called Patankar trick and guarantees non‐negativity of the water height for any time step size while still preserving conservativity. In the DG context, we prove consistency of the discretization as well as a truncation error of the third order away from the wet–dry transition. Because of the proposed modification, the implicit scheme can take full advantage of larger time steps and is able to beat explicit time stepping in terms of CPU time.
